Yeah, so the plot would have to be less murder-mystery, and more action-adventure/rom-com then.

The sleekness and opulence being contrasted with a grisly murder works with Orient Express.

So to get a contrast with the rugged and dirty and tiring road, you'd need the nobleness of the protagonist/s to shine above that.

That or you could have a really depressing tragedy as a road movie.
